How do you stain BrdU?
Detect incorporated BrdU
- Remove this solution and add 1 mL of antibody staining buffer.
- Add anti-BrdU primary antibody.
- Incubate overnight at room temperature.
- Wash with Triton X-100 permeabilization buffer (3 times, 2 minutes each)
- Add fluorescently labeled secondary antibody.
- Incubate one hour at room temperature.
What is BrdU positive?
BrdU-positive cells without co-labeling and BrdU/RCA-positive cells occur first after the H-I injury (significant increase at P10), followed by BrdU/NeuN-positive cells (significant increase at P42). Colabeling of BrdU and NeuN was confirmed by confocal microscopy (Figure 2).
